We have used CALTX SAE 40 in Gas Engine sump capacity of 750 Ltrs. During top up, one of the operator mix 35 Ltrs SAE 15W40. Considering the sump capacity is it required to drain all the lube oil ? is there any guideline for such contamination.

For such thing you need to do proper oil analysis to check the quality of oil. You can also use quality filtration carts to remove any kind of air or water contamination that can damage equipment/machine. A good oil and lubrication practice can only save you from such issues.
